One of the things I want to do is use some transitions like modal overlays and activity displays while pages load – in other words do some Ajax based UI functionality but with data that is provided by the host application. I am checking if user exists in database if exists I am showing message as 'existed user' and then I need to disable the signup button if not I need to enable it.
I am unable to enable and disable the sign Up button.
Can anyone help me in this issue?

Unfortunately your problem is something as small as the difference between
enabled
and disabled
.enabled = true;
Should be :
.disabled = false;

u can just set visibility of your button to false visibility='false'

The .disabled does work, have you used break point to make sure your code was even being reached ? Your conditional if / else's may not be returning your expected values.

To enable and disable the buttons using JavaScript, this is what should be done-

Enable or disable Javascript in Internet Explorer 11 using these easy steps.
- Select the Gear in the upper-right corner of the screen or the “Tools” menu if you have the menu bar enabled, then select “Internet Options“.
- Select the “Security” tab.
- Select the zone you wish to modify. In most cases, it will be “Internet“.
- Select the “Custom level…” button.
- Scroll down to the “Scripting” area and select the radio button to “Enable” or “Disable” it “Active scripting“. You may also opt for IE11 to “Prompt” you to allow scripts to run.
- Select “OK“, then “OK” again.
JavaScript should now be enabled or disabled in Internet Explorer 11.

Why is the “Custom Level…” button grayed out for me?
You may not have permission to modify settings in IE. Try closing IE, then re-open it by right-clicking the icon, then choosing “Run as Administrator”. If that doesn’t work, you may need to login as an admin to the system. If you are working in a corporate or school environment, you may need to contact your IT department.
